چکیده

The activation of NADPH oxidase on the plasma membranes of polymorphonuclear leukocytes (PMNL) follows the change of membrane potential. It is thought that the activation of NADPH oxidase is disturbed in the PMNL of patients with chronic granulomatous disease (CGD). The change of membrane potential was examined in the PMNL of normal subjects and patients with CGD, with a lipophilic probe, di-OC5(3). There was a disturbance in phorbol myristate acetate (PMA) and formyl-methionylleucyl-phenylalanine (FMLP)-induced depolarization but not in Ca ionophore A23187induced depolarization of the PMNL of patients with CGD. A23187-induced depolarization was not always associated with 0 2 release in normal PMNL. A23187 differed from PMA and FMLP in several aspects of its action on depolarization. It is concluded that only depolarization which couples with 0 2 release is disturbed in the PMNL of patients with CGD. Increase in extracellular potassium (K) caused PMNL of both normal subjects and CGD patients to depolarize but made them not to release 0 2-. Otherwise, A23187-induced depolarization was disturbed in PMNL of CGD patients with higher extracellular K concentrations. These findings help to confirm the theory that K efflux on the plasma membrane is disturbed in the PMNL of patients with CGD. 173

Arachidonic acid metabolism in polymorphonuclear leukocytes from patients with chronic granulomatous disease.

The effect of the calcium ionophore A23187 on the release and metabolism of [3H]arachidonic acid was examined in normal polymorphonuclear leukocytes and those obtained from patients with chronic granulomatous disease. The ionophore A23187 which stimulates oxidative metabolism in normal polymorphonuclear leukocytes was ineffective in increasing oxidative metabolism (chemiluminescence) in polymor...

Leukocyte degranulation and vacuole formation in patients with chronic granulomatous disease of childhood.

A cellular defect associated with decreased bactericidal activity of the polymorphonuclear leukocyte has been found in a 2(1/2) yr old Negro boy with the typical clinical and pathological findings of chronic granulomatous disease. Unlike previously described patients his polymorphonuclear leukocytes were shown to undergo apparently normal degranulation and vacuole formation after phagocytosis. ...

In vitro bactericidal capacity of human polymorphonuclear leukocytes: diminished activity in chronic granulomatous disease of childhood.

Diminished bactericidal capacity was found to be characteristic of polymorphonuclear leukocytes (PMN) from five children with the clinical syndrome of granulomatous disease of childhood. The PMN from these children demonstrated nearly normal phagocytic capacity, and the majority of viable bacteria, after 2 hours of incubation in the phagocytosis system, were found associated with leukocytes. Th...

X-linked inheritance in females with chronic granulomatous disease.

Chronic granulomatous disease in males is familial and its transmission is is usually clearly x-linked. The mode of inheritance in females with the syndrome is unknown and the carrier state difficult to identify. Defective polymorphonuclear leukocyte bactericidal activity in this disease is associated with an absence of the respiratory burst generated in stimulated phagocytes and may be detecte...

Studies of the metabolic activity of leukocytes from patients with a genetic abnormality of phagocytic function.

Polymorphonuclear leukocytes from patients with chronic granulomatous disease respond to the phagocytosis of latex particles with normal increments in glucose consumption, lactate production, Krebs' cycle activity, and lipid turnover. The leukocytes of these patients fail to show normal increments in respiration, direct oxidation of glucose, and hydrogen peroxide formation during particle uptak...
